According to the wiki, Tie Defenders are part of the Imperial Remnants. So you have to progress the main story until you reach that point(after the Battle of Endor).

Yeah they are still (I think) buildable at tech lvl 4/5 i.e. only after Endor and only after you clicked/built the research option after Endor (e.g. on Coruscant). When you get Thrawn he has a sqdr of those also I recall on the Chimera. I also think you may get them earlier as mission rewards.

In XPgamers ongoing YT playthrough he got them buildable earlier though i.e. before Endor, might be because he failed the Thrawn mission or he has a special build, not sure.

He has a older version where there was a bug where u could get IR tech without endor. This bug has been fixed for quite a while
