The buttons refer to the number of human players; so if you select 1 Player, it's "1 Human Player", 2 Player is "2 Human Players", etc. There will always be four players total, so if there are less than 4 human players, the game will fill the remaining slots with the AI.

Did you select 4-player mode? 4-player means "all humans" and thus no AI will spawn.

Otherwise, you may have an addon that is preventing them from functioning.
