Cleanse: So I can be lazy with vacuum. Also her attack is pretty nice too.
Dodge: To increase the the value of crystals throughout the run
Sparky: It's easy enough at the start (when you have no upgrades) to buff electricity to hit multiple mobs, as each upgrade makes it do double damage (on the upgrade tree).
I generally start with sparky and dodge, but put all levels into Dodge so he can start increasing that crystal value ASAP. Then level Sparky, and find Cleanse.
After that, I start to get Chop/Chill/Plague/Burny as they increase the damage taken by mobs.
IF you really want to min/max the Dodge Crystal buff, get Thor. Right before Thor's special procs, kick out everyone but Dodge, making Thor copy dodges buff. The guide says "Will copy and use the next special skill performed by a character in the cart.", so maybe there is an easier way to set it up so Thor always copies Dodge, but I'm not sure.
