Yes, you need to have the multimeter in the circuit on the diagram.

Either measuring R or I and V is acceptable... It just needs to fit your method, your diagram, and your materials list. I had expected I and V, but many people discovers the ohmmeter... just remember to explain how it is set up correctly in the circuit.
