I like how slow Optimus Prime's first transformation is. He's just essentially reshaped his entire body and is just taking his time, making sure all his parts go where they ought to and that they fit properly into place. Also, at various points you can hear hissing noises, as if air's being forced out as various parts come together.

Then, there's how he approaches Sam and Mikaela. Barricade (the Decepticon disguised as a police car) got right in Sam's face, addressing him as LadiesMan217- his eBay username.

Optimus Prime, by contrast, slowly transforms into robot mode, then slowly lowers himself down to look the two humans in the eye as best he can, and then addresses Sam by his actual name despite knowing Sam's eBay username. You can even see Optimus relax a little after Sam confirms that, indeed, they have found the right guy.
